چکیده

Title of Thesis: ENZYME INHIBITION IN MICROFLUIDICS FOR RE-ENGINEERING BACTERIAL SYNTHESIS PATHWAYS Dean Larios Berlin, Masters of Science, 2009 Directed By: Professor Gary W. Rubloff Department of Materials Science and Engineering and the Institute for Systems Research Enzyme-functionalized biological microfluidic (EF-BioMEMS) systems are an emerging class of lab-on-chip devices that manipulate enzymatic pathways by localizing reaction sites in a microfluidic network. An EF-BioMEM system was fabricated to demonstrate biochemical enzyme inhibition. Further, design optimizations to the EFBioMEM system have been proposed which improve system sensitivity and performance. The pfs enzyme is part of the quorum-sensing pathway that ultimately produces the bacterial signaling molecule AI-2. An EF-BioMEM system was fabricated to investigate the pfs conversion activity in the presence of a transition state analogue inhibitor. A reduction in enzyme conversion was measured in microfluidics for increasing inhibitor concentration that was comparable to the response expected on a larger scale. This EF-BioMEMS testbed is capable of investigating other compounds that inhibit quorum sensing. Design improvements were demonstrates that improve overall system responsiveness by minimizing unintended reactions from non-specifically bound enzyme. EF-BioMEMS signal-to-background performance increased from 0.72 to 2.43. ENZYME INHIBITION IN MICROFLUIDICS FOR RE-ENGINEERING BACTERIAL SYNTHESIS PATHWAYS

بیوسنتز نانوذرات نقره با استفاده از باکتری اشریشیا کولای

Background: Today, the synthesis of silver nanoparticles is very common due to their many applications in various fields. The synthesis of these nanoparticles is done by means of physical, chemical, or biological methods. However, due to its inexpensive and environmentally friendly features, the biological method is more preferable than the other two methods. Previous research has shown that ni...

Mechanism-Based Studies of the Active Site-Directed Inhibition and Activation of Enzyme Transketolase

Derivatives of phenyl-keto butenoic acids have been reported to be inhibitors of pyruvate decarboxylase, (PDC). The inhibition of transketolase, a thiamine requiring enzyme such as PDF, by meta nitrophenyl derivative of 2-oxo-3-butenoic acid (MNPB) is reported here. These studies indicate that the inhibitor binds to the enzyme at the active site. A two-step inhibition was observed, first th...

Inhibitory Effects of Swietenia Mahagoni Seeds Extract on Α-Glucosidase and Α-amylase

This study analyzed the inhibition activity of Swietenia mahagoni seeds extract on α-glucosidase and α-amylase enzymes inhibition assays.  Swietenia mahagoni seeds were extracted by using Supercritical Carbon Dioxide (SC-CO2) extraction at pressures of 20- 30 MPa and temperatures of 40- 60°C.  The oil yields obtained were analyzed with α- glucosidase and α- amylase enzymes inhibition assays.  A...
